The 23-year-old South Korean skater posted a score of 74.92, while Russia's Adelina Sotnikova sits second on 74.64.
Fifteen-year-old Yulia Lipnitskaya, who won gold in the team competition for the host nation in the opening weekend of the Games, stumbled during her routine, leaving her fifth.
Meanwhile, Great Britain's Jenna McCorkell, who finished 29th in Vancouver, is 25th.
The free skating performances will take place tomorrow.
