Anwar El Ghazi finally mustered Ajax's first shot on target with around 15 minutes remaining, but his effort was comfortably saved as Frank de Boer's side continued to struggle to create anything of note.
With around five minutes remaining, substitute Djurdjevic netted with the winner with a skilful effort, taking a cross on his chest before flicking the ball past a defender and rifling a half-volley past Cillenssen.
The win leaves PSV Eindhoven with a nine-point lead at the top of the table.
