The Argentina international scored late on to seal a 4-0 victory at Estadio de Son Moix as Barca stretched their lead at the top to five points.
In finding the net, the 32-year-old became the only player to hit the 20-goal mark in 12 successive league campaigns, a run that started with his 23-goal haul in 2008-09.
Messi also set up goals for Martin Braithwaite and Jordi Alba after Arturo Vidal had given the Catalan giants an early lead.
It was Barca's first game back following a three-month hiatus owing to the coronavirus pandemic.
