Both teams entered the game at the Cotton Bowl requiring a win after losing their opening fixtures, but a second-half goal from Francesco Totti was enough to earn victory for the Serie A side.
Real, who won the Champions League last season, dominated the opening 45 minutes, with Gareth Bale going close on more than one occasion.
However, 13 minutes after the restart, Alessandro Florenzi made inroads down the left before crossing for Totti to provide the first-time finish from 12 yards.
The conclusion of the match was marred by a number of supporters invading the pitch before Roma confirmed their win.
