Dani Carvajal, who has been advised to spend time off due to a heart condition, drops out in defence along with forward Gareth Bale, who is not included in the squad due to muscle fatigue.
Academy full-back Achraf Hakimi, 18, is in line to make his first start for the club, while Marco Asensio comes in for Bale.
As for Espanyol, Pablo Piatti misses out, which sees Marc Roca come into a team which includes both Gerard Moreno and Leo Baptistao in the final third.
In addition, Javi Fuego comes in for Papakouly Diop, while Sergio Garcia starts on the bench once again.
