The world number 107 hits six birdies and an eagle as he opened up a one-shot advantage over Charl Schwartzel and England's Chris Hanson.
George Coetzee and Keith Horne are a shot further adrift in a share of fourth position, while Branden Grace features in a group of eight players who are all three shots behind Stone.
British representatives Scott Jamieson, Richard McEvoy and Nathan Kimsey are also in contention in Madelane, as is Ireland's Paul Dunne who holds a place inside the top 20.
Spain's Alvaro Quiros - who has dropped down to 580 in the world rankings - has missed the cut after following Thursday's 81 with a round of 74.
