The tight end has missed the last two practices with the team, and is not expected to feature in the team's pre-season game against the Buffalo Bills on Sunday.
The 23-year-old reported pain in the foot during the Colts' rest day on Wednesday, initially seeking treatment at the team's practice facility.
Justice Cunningham, who was the final pick in this year's draft, is expected to split the first-team reps with Coby Fleener in Allen's absence.
