The visitors had to contend with an early setback as Rhys Williams was forced off within the first three minutes, Milos Veljkovic taking his place on the pitch.
Boro had the first concrete chance of the game on the quarter-hour mark, Jelle Vossen just failing to connect with a James Husband cross, before Kike headed wide.
Several minutes later, Vossen fired over the bar following good play by Patrick Bamford, before the hosts countered with a Conor Hourihane shot that also flew over the top.
Aitor Karanka's charges turned the screw as the game went on and saw a multitude of chances come close from the likes of Lee Tomlin and Daniel Ayala, but Barnsley were able to survive until the break.
