The visitors had the better of the early opportunities, with Antonio Amaya heading wide inside the opening 10 minutes, before Jorge Molina saw his strike saved.
Valladolid's first attempt on goal came in the 33rd minute when Daniel Larsson tried a long-range effort, but it went straight into the hands of Guillermo Sara.
The hosts made a strong start to the second half, as Javi Guerra and Fausto Rossi both came close, before just missing the net.
With stoppage time approaching, Betis applied pressure, with Juan Carlos almost squeezing it in from the edge of the box.
Guerra tried an optimistic shot in the final seconds, volleying the ball towards the away goal deep into injury time, but neither side could find the target before full time.
