First-half goals from Kenny Miller and Anthony Gerrard earned the Bluebirds a cup upset as they progressed into the last four.
Scotland international Miller put the home side in front when he latched onto Aron Gunnarsson's through ball and finished calmly through the legs of deputy Rovers keeper Mark Bunn.
Chances for Blackburn were limited in the opening periods, with David Dunn wasting their best opportunity by blazing over the bar.
Anthony Gerrard then doubled Cardiff's lead by heading in at the back post after Bunn failed to clear a Peter Whittingham corner.
Kean introduced Ruben Rochina, who scored in Rovers defeat to Stoke last weekend, but Cardiff hung on comfortably to secure a place in the Carling Cup semi-finals.
