That sparked Roma into life, as Francesco Totti went close to restoring parity before the half-time interval when he forced a good save out of Gianluca Pegolo.
The Lupi continued to dominate proceedings in Siena and deservedly equalised in the 63rd minute when former Siena striker Destro headed home Alessandro Florenzi's cross.
Roma went 2-1 up in the 86th minute as Simone Perrotta fired home from the edge of the Siena penalty box after picking up a loose ball from Matteo Rubin.
Then in the first minute of injury time Destro wrapped up the points for Roma when he shot in Miralem Pjanic's cross with his left foot.
