The duo retained the crown that they won at the Vancouver Games four years ago with a total time of 3:50.61.
United States duo Elana Meyers and Lauryn Williams saw their hopes of winning gold dashed during their final run, when they took a big hit early on and could not recover. They finished 0.10 seconds slower than Humphries and Moyse to take silver.
Americans Jamie Greubel and Aja Evans took the bronze medal while Esme Kamphuis and Judith Vis finished fourth for the Netherlands.
Great Britain's Paula Walker and Rebekah Wilson finished 12th with a time of 3:54.24.
