The Jaipur franchise will now face Mumbai Indians in Kolkata on Friday, with the winners going on to meet Chennai Super Kings in a title-deciding match two days later.
Brad Hodge was the hero with the bat for the Royals, hitting 54 not out off 29 balls as his side chased down their target of 133 with four balls to spare.
The 38-year-old Australian's impressive knock included two fours and five sixes, two of which came off Darren Sammy's bowling in the final over.
Hyderabad, who won the toss, had earlier made 132-7, with Shikhar Dhawan (33), Cameron White (31) and Sammy (29) doing the majority of the scoring.
