Leverkusen were given an early scare by the visitors when a loose ball dropped to Tarik Elyounoussi in the box, but Bernd Leno reacted impressively to make a fine save.
Leno was forced into action once again when Anthony Modeste sprinted clear of the home defence before unleashing a low effort which needed pushing to safety.
Hoffenheim goalkeeper Oliver Baumann matched the efforts of his opposite number by getting across his line impressively to keep out Calhanoglu's free kick.
A clever piece of skill from Heung-Min Son earned him space on the edge of the box after a patient move from the hosts, but he was left frustrated when Baumann got a strong hand to his low strike.
After making a fine stop to keep out a free kick earlier in the first half, Baumann was left helpless in stoppage time when Calhanoglu picked out the top corner with an unstoppable strike.
