Francesco Totti's fierce free kick from 35 yards was parried by Mattia Perin to the path of the 21-year-old, who tapped in with ease.
Totti forced the Pescara goalkeeper into a good save with another venomous free kick, before Roma goalkeeper Mauro Goicoechea almost let Francesco Modesto in.
Roma continued to control proceedings at Stadio Adriatico, as Destro's header from a Totti cross was brilliantly saved by Perin on the half-hour mark.
Then two minutes into the second half Alessandro Florenzi shot just wide from outside the penalty box.
Destro had two chances in quick succession moments later to add to his tally, but failed on both occasions.
Totti tried his luck from outside the Pescara area in the 57th minute but in vain, before Destro failed to keep his header from a Federico Balzaretti cross on target.
Totti was again denied by Perin on 76 minutes, and seven minutes later Marquinho latched onto Pablo Osvaldo's through-ball only to shoot wide.
