Adams gave the Blades the lead after just three minutes, firing in a rebound after Chesterfield keeper Tommy Lee parried a Conor Sammon effort.
Billy Sharp shot just wide in the eighth minute as the Blades looked to increase their lead.
A 31st-minute effort from Chesterfield captain Sam Morsy was kept out by United keeper Mark Howard.
Howard made another save from debutant Lee Novak before the end of the half.
Adams increased United's lead straight away in the second half, slotting low past Lee on 46 minutes.
Republic of Ireland international Sammon kept causing problems for the away side, and his cross saw a Stefan Scougall shot saved by Lee.
Sammon, making his home debut, had an effort disallowed after Lee was fouled during the initial United attack.
Adams' brace handed Nigel Adkins side all three points for their first league victory of the season.
