Oosthuizen hit seven birdies in a row on holes four to 10 as part of a miraculous run on the way to a stunning round of 63, which took him to a tournament score of 19-under par.
Northern Ireland's Rory Mcllroy lies three shots back on 16-under par after four birdies on the back nine ensured he remained in contention.
Tiger Woods and Dustin Johnson are both on 13-under par after they shot rounds of 65 and 68 respectively to give themselves an outside chance of challenging Oosthuizen at the top of the leaderboard.
